3.1.2. Tinh sch lipase bng phng php sc k trao i ion-DEAE-
cellulose trn thit b FPLC
Dch enzym sau ta bng etanol c ly 15 ml cho vo ct
c cn bng trong m Tris-HCl 25mM pH 7,5; tc chy l
1,5ml/pht. Dung dch y lipase l NaCl vi gradien nng 0-0,25M;
mi phn on thu 1ml/ng.

Theo hnh 3 cho thy khi nhit tng t 60C tr ln hot
lipase c nh tng dn v t cc i ti 70C (15 U/g). i vi lipase
tan, nhit 60C hot lipase t cc i (29,7U/g) v gim dn
khi nhit tng ln.
Nh vy nhit thch hp cho lipase t do v c nh hot ng
l khc nhau. Nhit ti u ca lipase c nh cao hn ca lipase t do
