Ngy nay enzyme c nh c s dng rt rng ri trong cc ngnh cng nghip
nh vo kh nng ti s dng v kh nng iu khin qu trnh sn xut bng enzyme.
Nghin cu ny kho st kh nng c nh Termamyl 120L trn silica-chitosan v silicaalginate. Enzyme c nh trn silica-chitosan c nhit v pH ti u ln lt l 60oC,
pH 8 v 60oC, pH 5,5 i vi enzyme c nh trn silica-alginate. Kh nng ti s dng
ca -amylase c nh trn silica-alginate (42 ln) cao hn kh nng ti s dng ca amylase c nh trn silica-chitosan (30 ln). S ng c nh glucoamylase v -amylase
mang li hiu qu cao v c th ti s dng ln ti 44 ln 50oC, pH 5,5. Cc phn ng
c thc hin trong 30 pht, iu kin nhit v pH thch hp.

ABSTRACT
Nowadays the immobilized enzyme used universally in industrials thanked to
capability re-use and industrial process control capability by enzyme. In this
research, a thermostable -amylase was immobilized by covalent bond onto silicachitosan beads and entrapment in silica-alginate beads. The optimum temperature
and pH of the enzyme -amylase, immobilized by covalent bond onto silicachitosan are 60oC, pH 8, respectively. For the immobilization enzyme -amylase
entrapped in silica-alginate, the optimum temperature and pH are 50 oC and pH 5,5,
respectively. The capability re-use of the immobilized enzyme -amylase entrapped
into silica-alginate beads (42 times) was higher than the immobilized enzyme onto
silica-chitosan beads (30 times). Co-immobilization glucoamylase and -amylase in
silica-alginate beads resulted in high yields and could re-use up to 44 times at 50oC
and pH 5,5. The reactions were performed within 30 minutes, at suitable
temperature and pH value.

2.1.
Vt liu c nh
Theo Trevan mt cht mang l tng cn c nhng tnh cht sau:

2.4.1. -amylase
-amylase c kh nng phn ct cc lin kt -1,4-glucoside nm pha trong phn

2.4.1.1. c tnh
Trung tm hot ng ca -amylase c cha nhm [COOH] v [NH2]. -amylase
d tan trong nc, trong cc dung dch mui v ru long.

Ngi ta cho rng c tnh ny ca -amylase c lin quan n hm lng ca Calci
trong phn t ca n (-amylase ca cc vi khun a nhit c cha Calci nhiu hn
amylase ca nm mc 3-4 ln nn n bn nhit hn). Tt c nhng -amylase u b

2.4.1.2. Gii thiu v enzyme Termamyl

thy phn lin kt 1,4 -glucoside thnh amylose v amylopectin.
Mt s ng dng ca Termamyl trong cng nghip (thng tin do hng Novo cung
cp) [38]:
- Trong k ngh tinh bt: Termamyl c dng cho vic dch ha lin tc tinh
bt trong ni hi hoc trong nhng thit b tng t hot ng nhit t 105 110oC v v vy li dng c tnh n nh nhit cao ca enzyme.

[34]. Phn ng c thc hin trong 8 gi v sau ht gel c ra sch bng

kh nng ti s dng c 20 ln vi tng lng thi gian phn ng l 160 gi. Mt
