1.2.4 Gii thiu enzyme pectinase
Enzyme pectinase l enzyme xc tc s phn hy ca cc polymer pectin. S
phn hy pectin trong t nhin thng xy ra khi tri cy chn. Nhng enzyme ny
v vy c mt vai tr ht sc quan trng trong qu trnh bo qun tri cy v rau

Polygalacturonase cn c tn gi l poly 1,4-galacturoniglucanohydrolase,

1.2.4.3 ng dng pectinase

1.3.1 Ch phm Biopep-21
c sn xut t chng Aspergillus niger. Chng vi sinh vt ny c cung
cp t phng th nghim Khoa Cng ngh Sinh hc, trng i hc Quc t.
Apergillus niger thng thy trn lng thc bo qun, lc va mi phn lp
thng c u nht nhy. Tuy nhin, tip tc nui cy, tnh cht ny bin mt

2.2.2 Phng php c hc
2.2.2.1. Kim tra cht rn ha tan chit xut t ht tiu
Ht tiu c nghin trong 5 pht bng my xay sinh t, v sau cn
10g ht tiu bt cho vo Erlen 250ml. Thm 110ml nc ct vo Erlen v lc trong
1 pht trch xut cc cht rn ha tan t tiu bt. Ly dung dch em lc qua giy
lc v cho 1 git dung dch lc ca ht tiu c lc a vo l trn khc x k
kim tra cht rn ha tan t ht tiu. [29]
2.2.2.2. Kim tra pH ca dung dch ht tiu
Ht tiu c nghin trong 5 pht bng my xay sinh t, v sau cn
10g ht tiu bt cho vo Erlen 250ml. Thm 110ml nc ct vo Erlen v lc trong
1 pht trch xut cc cht rn ha tan t tiu bt. Ly dung dch em lc qua giy

Cho 50g ht tiu ht tiu c ngm vi thi gian ph hp c c

2.2.3 Phng php ha sinh

2.2.5.1 Kho st m cho qu trnh ln men
Trc khi ln men ngm ht tiu vi thi gian khc nhau v kim tra m.
Cc ht tiu c chuyn sang r nha ro nc. Chuyn tt c tiu vi nhng
m khc nhau vo cc khn ln men v trn u vi mi mu 28% ch phm
sinh hc v ln men trong 48 gi. Sau 48 gi, ly cc mu tiu ln men ra sch
(xem 2.2.1.2) kim tra kh nng bc v v tin hnh phn cp (xem 2.2.1.3). Tnh
s lng ht tiu mi cp v tin hnh so snh gia cc mu c m khc nhau.

2.2.5.2 Kho st t l ch phm sinh hc cho qu trnh ln men
Ht tiu c ngm t c m tt nht cho qu trnh ln men.
Sau chng c chuyn n cc r nha ro nc khong 5 pht. Tin hnh
ln men vi cc t l ch phm khc nhau, trn u ch phm vi ht tiu. Tin hnh
ln men trong 48 gi. Sau 48 gi ln men ly mu ra loi b v ht tiu (xem

Khi b sung ch phm t, nm tng trng v pht trin tt nhng lng
enzym pectinase v cellulase khng bc v ht tiu hon ton v tt nht. Khi
t l ch phm c b sung qu nhiu, nm pht trin nhiu trn b mt ht tiu ly
cht dinh dng cho s sinh trng v pht trin ca nm, s khng cung cp
cho nm pht trin. Mt khc, m thch hp ln men b gim nhiu do nm s

3.5. Kim tra gi tr pH v cht rn ha tan ca ht tiu ln men
Gi tr pH ca ht tiu ln men thay i trong khong 6,0-6,55 (bng 3.3).
Tiu ln men bng ch phm Biopep-21, Biopep-22 gi tr pH tng nh v sau
gim dn. Sau 3 ngy tiu trng Biopep-21 c pH= 6,51, Biopep-22 c gi tr pH=
6,39. Tuy nhin gi tr pH t 6 n gn trung tnh khng nh hng n sc khe
ca con ngi v hng v cc mn n.
