Mc ch ca nghin cu l xc nh nhng chng x khun c tnh i khng cao vi nm hi thc vt. T 8 mu


t khc nhau, phn lp v lm thun c 43 chng x khun, phn thnh 7 nhm mu vi t l khc nhau: trng 37,2%; xm - 22,9%; nu - 14,0%; vng - 11,6%; hng - 2,3%; tm - 2,3%; xanh - 4,7%. Trong s c 16 chng i
khng nm Fusarium oxysporum, 11 chng i khng nm Botryosphaeria dothidea; 3 chng i khng nm
Phytophthora capsici v 4 chng i khng nm Sclerotium hydrophylum. tuyn chn c 2 chng x khun HN6
v NA1 c hot tnh khng nm mnh nht. Kho st kh nng sinh trng trn mi trng Gause I cho thy HN6 v
NA1 c kh nng sinh trng tt nhit 30C, pH trung tnh, ng thi, chng c kh nng chu nhit , nng
mui tng i cao. Kho st kh nng sinh trng trn mi trng ISP 9 cho thy HN6 v NA1 c kh nng s dng
cc ngun ng khc nhau: D - glucose, saccarose, D - xylose, rhamnose, raffinose. Bc u xc nh c
chng HN6 thuc loi Streptomyces roseosporus, chng NA1 thuc loi Streptomyces albofaciens.

Isolation and Screening Streptomyces spp. against Plant Pathogenic Fungi


ABSTRACT
The aim of the present study was to identify Streptomyces spp. antagonistic to plant pathogenic fungi. From
eight of different soil samples, a total of 43 isolates of Streptomyces were isolated and they were divided into 7
groups with different proportion: white 37.2 %; grey 22.9 %; brown 14.0 %; yellow 11.6 %; pink 2.3 %; purple 2.3 %;
and blue 4.7 %. Of 43 isolates, 16 were antagonistic to Fusarium oxysporum, 11 were antagonistic to Botryosphaeria
dothidea, 3 were antagonistic to Phytophthora capsici, and 4 were antagonistic to Sclerotium hydrophylum. Two
streptomyces isolates, HN6 and NA1 were selected due to their high fungal antagonism. HN6 and NA1 grew well on
Gause I medium at 30C, neutral pH and fairly high salt concentration. On ISP 9 culture medium HN6 and NA1 were
able to utilize various sugar sources: D - glucose; saccarose, D - xylose, rhamnose and raffinose. The isolate HN6
was preliminarily identified as Streptomyces roseosporus, while NA1 as Streptomyces albofaciens.

2. VT LIU V PHNG PHP


2.1. Phn lp x khun v chun b mu
nm bnh cy
Mu t c ly su t 5-20cm ti cc
a im H Ni, Hng Yn, Nng, Vnh
Phc, Bc Giang, Thi Bnh, Ngh An v Gia
Lai. X khun c phn lp theo phng php
ca Vinogradkii (1952) (trch theo Nguyn
Thnh t, 2000)
Cc loi nm bnh cy bao gm: Fusarium
oxysporum, Phytophthora capsici, Botryosphaeria
dothidea v Sclerotium hydrophylum c cung
cp bi Trung tm Bnh cy Nhit i - Hc vin
Nng nghip Vit Nam.
2.2. Kho st cc chng x khun c kh
nng i khng nm
kho st kh nng i khng ca cc
chng x khun vi cc mu nm bnh cy,
nghin cu s dng 3 phng php nh sau:

mi trng Potato Dextrose Agar (PDA - g/l:
Khoai ty ming 200, ng 20, NaCl 20, Agar
20, pH = 7,4). Nm bnh c cy trung tm
a petri, chng x khun c cy 4 gc bao
quanh nm bnh cch tm a 3cm. ng knh
vng c ch sinh trng c xc nh sau 4 - 7
ngy nui cy 30oC.
- Phng php khuch tn a thch
(Dhanasekaran et al., 2012): X khun c
nui trong mi trng Gause I lng (g/l: Tinh
bt tan 20g; K2HPO4 0,5g; MgSO4.7H2O 0,5g;

NaCl 0,5g; KNO3 0,5g; FeSO4 0,01g; pH =7,4),


lc 200 vng/ pht 30oC. Dch x khun c
thu sau 7 ngy nui cy. Nm c hot ha v
lm thun trn mi trng PDA, dng que cy
ly si nm cho vo ng eppendorf cha 500l
nc ct v trng, voltex bo t nm pht
tn u trong nc. Ging thch c to trn
a thch PDA c cy tri 50l dung dch
nm. 100l dch x khun c ly tm vi tc
8.000 vng/ pht trong 30 pht, 4oC v dch x
khun khng ly tm c b sung vo ging
thch, 30oC. Dch x khun c kh nng c
ch sinh trng ca nm c th hin thng
qua vng sng xut hin quanh ging thch.
- Phng php thi thch (Nguyn Ln
Dng v Phm Th Trn Chu, 1978): X khun
c cy u trn a petri cha mi trng
Gause I 30oC. Sau 7 ngy nui cy, thi thch
x khun c cy vo a petri cha mi
trng PDA c cy tri nm, 4oC trong
4 - 5 gi cc hot cht t thi thch khuch
tn vo mi trng, sau cho vo t nui,
quan st kt qu sau 4 ngy.
2.3. Nghin cu mt s c im sinh hc
ca cc chng x khun c kh nng
khng nm bnh cy
c im hnh thi ca chng x khun
c xc nh da trn cc c im nui cy bao
gm: mu sc ca khun ty kh sinh; mu sc
ca khun ty c cht; kh nng sinh sc t tan
(Tresner and Backus, 1963) v s hnh thnh sc
t melanin trn mi trng GauseI, GauseII (g/lNc chit tht 30ml, Pepton 5, NaCl 5, Glucose
10, Agar 20) v h thng mi trng ISP (ISP1
(g/l- Tryptone 5, cao nm men 3, Agar 20, pH =
7.0); ISP2 (g/l- Cao nm men 4, dch chit Malt
10, glucose 4, Agar 20, pH = 7,3); ISP3 (g/l- Bt
yn mch 20, Agar 20, dung dch mui vi lng
1,0 ml, pH = 7.0); ISP4 (g/l- Tinh bt 10, K2HPO4
1, MgSO4.7H2O 1, NaCl 1, (NH4)2SO4 2, CaCO3 2,
dung dch mui vi lng 1.0 ml, Agar 20, pH =
7,0); ISP5 (g/l- L - asparagin 1; glyxerin 10;
K2HPO4 1; dung dch mui vi lng 1,0 ml; agar
20, pH = 7,0); ISP6 (g/l- Peptone 10, cao nm
men 1, xitrat st 0,5, agar 20, pH = 7.0); ISP7
(g/l- Glycerin 15; L - tyrosin 0,5; L - asparagin 1;

K2HPO4 0,5; MgSO4.7H2O 0,5; NaCl 0,5;


FeSO4.7H2O 0,01; dung dch mui vi lng 1,0
ml, agar 20, pH = 7,0).

3. KT QU V THO LUN
3.1. Phn lp v tuyn chn cc chng x
khun

X khun c nui cy trn mi trng


ISP9 (g/l- (NH4)2SO4 2.64; KH2PO4 2.38;
K2HPO4.3H2O 5,65; MgSO4.7H2O 1; dung dch B
1,0 ml; agar 20; pH = 7,0) c b sung 1% cc
ngun ng khc nhau nh: D-glucose; Larabinose; saccarose; D-xylose; I-inositol;
mannitol; rhamnose; raffinose; cellulose; lactose
xc nh kh nng ng ha cacbon ca chng
thu c. Dung dch mui B (%): CuSO4.5H2O
0,64; FeSO4.7H2O 0,11; MnCl2.4H2O 0,79;
ZnSO4.7H2O 0,15; nc ct 100ml.

t, x khun chim khong 20 - 40% tng s vi
sinh vt trong t, tp trung nhiu lp t b
mt (Mitra et al., 2008; Proudyogiki, 2012). T
cc mu t khc nhau, 43 chng x khun
c phn lp trn mi trng Gause I vi cc
c im hnh thi v mu sc khun lc khc
nhau. Mu sc ca h si kh sinh c xc nh
da vo bng mu ca Tresner v Buckus. Cn
c vo mu sc khun ty kh sinh, cc chng x
khun c chia thnh 8 nhm mu khc nhau
(L Gia Hy, 1994; Trerner and Buckus, 1963).
Trong s 43 chng x khun phn lp c c 7
nhm mu xut hin vi s lng v t l khc
nhau (Bng 1).

Kh nng sinh enzyme ngoi bo c xc

b sung c cht (Gulve and Deshmukh, 2011).
X khun c cy chm im trn mi trng
a thch Mineral salt agar (g/l- (NH4)2SO4 4;
NaCl 6; K2HPO4 1; MgSO4 0,1; CaCl2 0,1; c cht
(CMC, tinh bt, xylan) 10 hoc gelatin 0.1, Agar
20, pH = 7,0), sau 7 ngy nui, a petri mc
khun lc c em nhum bng thuc nhum
lugol 1% kim tra hot tnh amylase,
cellulose, xylanase v nhum bng amido en
10B 0,1% kim tra hot tnh protease. Hot
tnh enzyme ngoi bo c th hin qua vng
sng quanh khun lc x khun.

3.2. Kho st kh nng i khng nm

cht khng sinh (CKS) c ch s pht trin ca
cc loi nm gy hi thc vt v con ngi (Qin
et al., 1994). Chng c s dng trong qu trnh
ln men sn xut cc hp cht c tnh khng
nm, khng virus, chng ung th, thuc tr su,
thuc dit c. Sau khi phn lp v lm thun 43
chng x khun, tuyn chn cc chng x
khun c hot tnh khng nm cao, chng ti
tin hnh kim tra s b hot tnh khng nm
ca cc chng x khun bng cc phng php
ng nui cy, phng php khuch tn a
thch v phng php thi thch.

Nghin cu nh hng ca iu kin nui

Trong 43 chng x khun phn lp c, 16


chng c kh nng i khng nm Fusarium
oxysporum (chim 37,2%), 12 chng c kh
nng i khng nm Botryosphaeria dothidea
(chim 25,6%). Phn ln cc chng ny i
khng yu vi nm Phytophthora capsici v
Sclerotium hydrophylum. Trong s cc chng x
khun c kh nng i khng, chng ti chn
c hai chng x khun NA1 v HN6 c kh
nng i khng vi c 4 loi nm gy hi trn.
Chng ti tin hnh ng nui cy hai
chng x khun NA1 v HN6 vi nm bnh cy
trn mi trng Gause I v theo di sau bao lu

ca hai chng x khun ny. Chng NA1 duy
tr i khng vi 4 loi nm bnh lu hn so vi
chng HN6. Chng NA1 c th duy tr i
khng vi F. oxysporum, B. dothidea v S.
hydrophylum t 19 n 22 ngy, chng HN6
duy tr i khng nm bnh t 5 n 17 ngy.
Khi kho st kh nng i khng vi P. capsisi
chng ti thy chng NA1 duy tr i khng n
7 ngy, chng HN6 ch duy tr n 4 ngy. Sau
thi gian ny ti vng i khng thy xut
hin cc khun lc nm bnh.

3.3. Mt s c im sinh hc ca hai

Mu sc khun lc ca mt chng x khun

Chng NA1 khi nui cy c 7 ngy KTKS
c mu trng trn cc mi trng Gause I,
GauseII, ISP1, ISP2, ISP3, ISP4; mu trng
xm trn mi trng ISP5, ISP6, ISP7. Sau 21
ngy nui cy, KTKS c mu xm trn ISP2 v
ISP5, trn cc mi trng khc KTKS khng
thay i. Trn mi trng GauseII, t ISP2 n
ISP7, KTCC c mu vng khng thay i sau 21
ngy; trn Gause I v ISP1 KTCC c mu trng.
Khi nui cy trn cc mi trng Gause v
h thng mi trng ISP, c 2 chng x khun
HN6 v NA1 u khng lm thay i mu sc
mi trng, chng t chng khng c kh nng
sinh sc t tan v khng hnh thnh sc t
melanin.
3.3.2. c im sinh l, sinh ha

Chng HN6 ng ha tt nht: D - glucose;
saccarose; D - xylose; rhamnose; raffinose, sinh
trng yu trong mi trng: L - arabinose; I inositol; mannitol; cellulose; lactose. Chng
NA1 c kh nng ng ha tt: D - glucose;
saccarose; I - inositol; mannitol; raffinose;
lactose, sinh trng yu trong mi trng c
cha L - arabinose; D - xylose v khng c kh
nng ng ha 2 ngun ng rhamnose v
cellulose. Ngun cacbon c th c th c s
dng c hiu qu bi chng ny nhng khng
hiu qu bi chng khc cho thy ngun cacbon
c th ny c th khng phi l ngun cacbon
thch hp hay c cha thm mt lng rt nh
(traces) cc thnh phn khc nh Oskay et al.
(2004) cng b
Qu trnh sng v trao i cht ca vi sinh

nhit , pH v nng mui khc nhau. C hai
chng u sinh trng tt nht 30oC, thch
hp vi mi trng trung tnh hoc hi kim v
c kh nng chu mn, pht trin tt nht pH
= 7-8, v nng mui t 5-7%. Theo Larsen
(1986) vi sinh vt chu a mui c th nhm
thnh cc nhm theo nhu cu v mui ca
chng, cc sinh vt chu nng mui thp c
th sinh trng trong mi trng nc bin vi
nng mui t 2-3%. Cc sinh vt thuc nhm
chu mui trung bnh c th sinh trng ti
nng NaCl t 5-20% (w/v). Nhm sinh vt
chu nng mui cao c th sinh trng ti
nng mui bo ha, khng sinh trng khi
nng NaCl thp hn 12%. Cc chng x
khun thuc nghin cu ny chu nng mui
ti 7% nn c th xp vo nhm chu mui trung
bnh. Kt qu nghin cu ca chng ti ph
hp vi cc nghin cu cng b ca Nguyn
Th Minh Hng v Vn Bt (2013). c bit,
chng HN6 c kh nng chu nhit 45oC,
chng NA1 c kh nng chu nhit 40oC.

Khng phi tt c cc vi sinh vt u c kh


nng sinh enzyme nh nhau v ngay c nhng
chng trong cng mt loi cng khng c hot
tnh nh nhau. V vy, khi tuyn ging phi
tin hnh phn lp, kim tra v la chn cc
chng c hot tnh enzyme mnh, sinh nhiu
enzyme mong mun theo tng mc ch. Chng
ti tin hnh kim tra kh nng ny ca 2
chng x khun nghin cu. Kt qu trn cho
thy, chng HN6 c kh nng sinh c 3 loi
enzyme ngoi bo nhng mnh nht l
protease. Chng NA1 sinh amylase v cellulase
vi hot tnh cao nhng khng sinh protease
ngoi bo.

Trong qu trnh sng, x khun c kh


nng tit vo mi trng enzyme ngoi bo
phn gii cc hp cht hu c phc tp thnh
cc cht hu c n gin c th hp th c.

T kt qu so snh cc c im hnh thi,


sinh l, ha sinh, chng ti tm thi t tn
chng HN6 l S. roseosporus HN6 v t tn
chng NA1 l S. albofaciens NA1.

3.4. Kt qu phn loi chng HN6 v NA1


So snh c im phn loi ca chng HN6
v NA1 vi kha phn loi x khun theo Gause
(1983) chng ti nhn thy, chng HN6 c
nhiu c im ging vi loi x khun chun S.
roseosporus, chng NA1 ging vi loi S.
albofaciens (Bng 4).
