UV-Visible Spectroscopy

The principles and techniques of ultra-violet and visible spectrophotometric methods.

UV-Visible Spectroscopy (UV-VIS analysis) is part of the Spectrophotometric Lab Skills curriculum. Course Reference Code is 3001UV.

Goal
The goal of this module is to provide basic information about UVVisible Spectroscopy (UV-VIS analysis) and its importance in a pharmaceutical analytical testing laboratory.

3001UV-LO03: Given accurate visual examples of equipment components and tools of the equipment involved with UV-VIS analysis, identify their name, function and importance Light source Tungsten-halogen lamp Deuterium arc lamp Sample Cuvette Detector Single-beam instrument Double-beam instrument Spectrogram 3001UV-LO04: Describe, in general terms, the processes and techniques related to UV-VIS analysis, including: Advantages and disadvantages of a single-beamed instrument Advantages and disadvantages of a double-beamed instrument Cuvettes Performing a qualitative analysis Performing a quantitative analysis 3001UV-LO05: Identify the importance of UV-VIS analysis in the pharmaceutical industry and how it is used with respect to the safety, identity, strength, purity, and quality of products. 3001UV-LO06:Identify the safety practices and equipment that are intended to prevent injury to those in the laboratory and damage to equipment, instruments, and facilities. 3001UV-LO07: Given examples of techniques used to perform UV-VIS analysis, differentiate between acceptable (good) and unacceptable (poor) practices that involve the following: Worker health and safety Cuvette handling Cuvette cleaning Care and maintenance of the sample compartment Care, handling, and maintenance of source lamps Calculating the amount of light absorbed by the sample Calculating the purity from the concentration

3001UV-LO08: Identify the correct steps and sub-steps in the proper sequence for using or performing UV-VIS analysis. PREPARE Verify instrument and its calibration status Turn on instrument and allow to stabilize Select and clean the appropriate cuvette(s) Prepare sample/reference solution PERFORM Pre-rinse cuvette with sample/reference solution 3-5 times Fill cuvette with sample/reference solution Clean outside of cuvette Inspect Insert cuvette into instrument Record absorbance and other required data Remove cuvette Clean cuvette Clean instrument when finished Perform calculations as required for qualitative analysis Perform calculations as required for quantitative analysis 3001UV-LO09: Given a question related to UV-VIS analysis that could be asked by a new laboratory analyst, provide an answer that is consistent with standard industry and scientific practices.
